If a virus does get through, the brain does have defenses to slow the viruses from reproducing. Only four of the patients exhibited encephalitic signs, all infected by the Herpes Simplex virus. Samples taken from acyclovir-treated mice ten days post viral infection had virus quantities in the brain that were equal to or exceeded the control group, suggesting a rebound of viral replication in these tissues. Contusions can occur directly under the impact site (i.e, a coup injury) or, more often, on the complete opposite side of the brain from the impact (i.e., a contrecoup injury). CNS vascular permeability was induced as previously described (15, 20, 28). However, these genes are expressed in a variety of cell types in different tissues, and thus complete knockout of these transporters would globally change the localization of their substrates, and thus their specific function at the BBB may be obscured. This is explained by the restoration in function of the impaired blood-brain barrier that occurs as the inciting infectious agent is controlled.

Viral vectors are now routinely used as tools for studying gene function as well as developing gene-based therapies for a variety of diseases. The mechanism also includes carriers that transport necessary substances from the blood to the brain and removes waste products. This barrier may not be well-known, but it performs a very important function: keeping your brain free of damaging chemicals and microorganisms that may be circulating inside your bloodstream. In both in vitro (laboratory) and in vivo (animal) studies, low, non-toxic doses of hypericin significantly inhibited the replication of several viruses, including HIV, influenza A, cytomegalovirus (CMV), Herpes simplex 1 and 2 (HSV-1 and HSV-2), and Epstein-Barr virus (EBV). If these results can be reproduced in humans, they may facilitate gene therapy of diseases that affect large areas of the brain, such as motorneuron diseases, Parkinson’s disease and Alzheimer’s disease. These studies compared the efficacy of CMX001 to ACV in BALB/c mice inoculated intranasally with HSV types 1 or 2. To obtain data on its penetration across the blood-brain barrier, we determined concentrations of foscarnet in cerebrospinal fluid and in plasma specimens from 26 patients with human immunodeficiency virus (stages 2 to 6 by Walter Reed Army Institute of Research classification) after a single infusion of 90 mg of foscarnet per kg of body weight and at steady state by electrochemical detection by high-pressure liquid chromatography.
